Building Tumor-Activated Biologics Through a Partnership Designed to Grow

From early company formation to next-generation masked T-cell engagers, explore how Voro Therapeutics and Alloy Therapeutics are building a scalable partnership to advance tumor-activated biologics.

Building Tumor Activated Biologics Through a Partnership Designed to Grow

Many promising oncology therapies are constrained not by a lack of potency, but by where that potency is active.

When a therapeutic target is found in healthy tissue as well as tumors, treatment can affect more than the cancer it was designed to address. That can limit dosing, narrow the therapeutic window, and prevent otherwise promising approaches from reaching their full potential.

Voro Therapeutics was founded around a different approach. Its PrimeBody™ platform is designed to keep biologics inactive in circulation and healthy tissue, then activate them selectively within the tumor microenvironment.

Turning that idea into a development-ready platform required more than a single discovery technology. It required integrated scientific capabilities, development infrastructure, and a partnership that could evolve alongside the company.

From Company Formation to an Integrated Drug Discovery Partnership

Voro’s relationship with Alloy Therapeutics began before Voro existed as an independent company. Alloy’s affiliated venture studio, 82VS, initially recruited Voro founder Ugur Eskiocak as an Entrepreneur-in-Residence to support the studio’s immuno-oncology portfolio. As the concept for Voro took shape, that relationship expanded into company-building and drug discovery.

“Alloy has been a fantastic partner for us,” said Eskiocak, Co-Founder and CEO of Voro Therapeutics. “Having access to their cutting-edge platforms, scientists, bench space and business experts allowed us to efficiently develop our technology and obtain proof-of-concept data with an exceptionally lean team.”

The model gave Voro access to research and development capacity as well as operational, strategic, and legal support without requiring the company to build every function independently from the beginning.

Scientifically, the teams brought complementary strengths to the collaboration. Voro contributed its expertise in masking biology, protease-cleavable linker design, and conditional activation. Alloy contributed antibody discovery, engineering, and development capabilities that could be organized around Voro’s product strategy.

Advancing VOR-101 from Concept to Development Candidate

The first major proof point was VOR-101, Voro’s masked, protease-activated CD47 antibody.

In preclinical studies, VOR-101 demonstrated curative anti-tumor activity and a favorable tolerability profile alongside monoclonal antibody-like manufacturability. The program advanced from an early concept to a development candidate and has progressed into IND-enabling studies.

The broader significance extends beyond one molecule. VOR-101 helped establish a foundation for applying the PrimeBody™ platform across additional targets and therapeutic formats.

Expanding Tumor-Activated Biologics into Masked T-Cell Engagers

Voro and Alloy are now building on that foundation through a strategic research collaboration focused on masked T-cell engagers. The work combines Voro’s masking and linker technologies with Alloy’s antibody discovery and multispecific engineering capabilities, applying a tumor-selective activation strategy to a modality that has historically faced difficult safety margins.

For Alloy, the collaboration reflects its Product Development Organization model: integrated discovery, engineering, and development capabilities built around a partner’s program. For Voro, it provides a scalable way to expand the PrimeBody™ platform while maintaining a focused internal team.

Building a Scalable Partnership for Next-Generation Cancer Therapies

What began as an early company-building relationship has grown into a collaboration spanning masked biologics, multispecific antibodies, and next-generation T-cell engagers.
